获得条件性基因敲除小鼠后如何进行下一步繁殖?如何选择实验对照小鼠?


条件性基因敲除小鼠(也叫Flox 小鼠)是指在目的基因中含有成对的loxp 位点的小鼠,与Cre 工具小鼠交配后可在

特定的组织或细胞中敲除目的基因。

方案一

若Flox 小鼠与广泛表达或生殖细胞特异Cre 小鼠交配,则可以获得全身基因敲除的KO 小鼠,繁育流程如图所示。ES

打靶载体中筛选基因neo 在两个loxp 之间,不需要单独去neo,结构为5’ arm-loxp-flox-frt-neo-frt-loxp-3’ arm,直

接配生殖腺表达的Cre 可同时去除neo 基因。

方案二

若与组织特异性Cre 小鼠交配,则可以按照图4 所示流程进行繁殖,一般采用转入Cre 重组酶的flox 杂合子小鼠与

flox 杂合子小鼠交配的方式获得Cre 酶整合的flox 纯合子条件基因敲除小鼠。此时,一般选择同窝出生的Cre 酶未整

合的flox 纯合子小鼠作为实验对照组。

方案三

也可以采用图5 的方案提高实验组小鼠比例。
